Samples can be submitted in two formats, either as
a gel slice or spot, or as a solution of proteins. There are specific
requirements for both formats, but a few requirements are general:
- The sample amount should be
more than 100 fmole
- The genome of the species from
which the sample originates has to be sequenced and annotated
- No
radioactive
samples !
- Sample volume smaller than
100 ul

Requirements for a gel slice or spot
|
- Gel can be stained with
either coomassie, colloidal coomassie or silver staining. In case of
silver staining, the staining has to be performed according to our
protocol (look at section protocols).
- Be careful for contamination
with keratins from skin or hair. Wear gloves, never touch the gel with
bare hands and use clean glass plates and scalpels for excision of
bands.
- Gel bands or spots can be
submitted in a simple eppendorf tube. The band can be stored dry at
-20 ºC.
- A picture of the gel from
which the band is excised should be included. Please circle the bands of interest.

Requirements for a solution of
proteins
|
|
The requirements are depending on the goal of the
experiment.

Requirements for in solution
digestion
|
- The sample should be absolutely free of
detergents
- Salt concentration should be < 500 mM
- Glycerol concentration should be < 2,5%
- No peptide elution
- No protease inhibitors
- No organic solvents
- Sample is preferably solved
in 8M ureum/thioureum
- Sample can be submitted in
dry form, either freeze dried of vacuum dried

Requirements for fractionation on 1D
gel electrophoresis
|
- Salt concentration should be < 500 mM
- Ureum concentration should
be < 0,5 M
